香蕉腺苷酸核糖基化作用因子1(MaArf1)基因的克隆及序列结构分析Cloning and structural characterization of ADP-ribosylation-factor 1 in banana (Musa accuminata L. AAA)

摘要(Abstract):
Arf(ADP-ribosylation factors)作为小G蛋白家族中的一类,在植物细胞的胞内运输中起着重要的作用。从采后2d的香蕉(Musa accuminata L.AAA group)果实cDNA文库中筛选到一条Arf基因,命名为MaArf1。序列分析表明,MaArf1在植物中是高度保守的。与水稻、苜蓿、马铃薯、小麦和拟南芥等物种中Arf基因的相似性都超过95%。并且MaArf1分别在24-31(序列为GLDAAGKT,Arf蛋白P结构域),67-71(序列为DVGGQ,G'结构域),126-130(序列为NKQDL,G结构域)的氨基酸处也具有保守的GTP结合结构域。根据已获得的cDNA序列设计引物,研究又获得了该基因的基因组序列。对其结构分析发现,该序列全长1998bp,由5个外显子和4个内含子组成。内含子序列中存在大量的重复序列,并且在第4个内含子中有一个完整的开放阅读框,推测这些序列可能参与基因的表达调控,也可能与RNA水平上的可变剪接有关。
