黄肉桃果实中类胡萝卜素提取和测定方法研究Extraction and analytical methods of carotenoids in fruit of yellow flesh peach

摘要(Abstract):
【目的】建立适于黄肉桃果实中类胡萝卜素有效提取和高效液相色谱(HPLC)快速测定的方法。【方法】在采用YMC-C30色谱柱,检测波长450 nm和柱温25℃的条件下,重点从提取料液比、提取方式和时间、液相测定的流动相配比、流速和进样量等方面研究了黄肉桃果实中类胡萝卜素提取和测定的方法,并对方法进行了评价。【结果】黄肉桃果实中类胡萝卜素以丙酮为提取剂,果肉与提取剂的料液比(g·m L~(-1))为1∶6,超声波避光辅助提取1 h;HPLC测定进样量20μL,以甲基叔丁基醚和甲醇(V/V=30∶70)为流动相等度洗脱,流速1.0 m L·min~(-1)。本方法操作简单,类胡萝卜素提取效率高,HPLC测定回收率为95~105%,相对标准偏差(RSD)小于2.25%,稳定性良好,精密度高,用时短,成本低。【结论】优化所得方法适于黄肉桃果实中类胡萝卜素的有效提取和快速测定。
