香蕉MYB基因克隆和表达分析Molecular cloning and expression of MYB1 gene from banana(Musa acuminata L.AAA group,‘Cavendish')under the various stresses

摘要(Abstract):
【目的】获得香蕉抗逆相关转录因子。【方法】通过随机克隆测序的方法从香蕉根系c DNA文库中获得MYB基因,命名为Ma MYB1。【结果】对扩增获得的c DNA序列进行生物信息学分析,表明Ma MYB1是香蕉MYB基因编码框全长c DNA,包含一个951 bp的最大开放阅读框,编码一个长316个氨基酸的蛋白质。蛋白质序列同源比对发现其含有完整的2个保守的MYB结构域R2和R3,属于典型的R2R3-MYB基因。系统进化树比对分析表明,Ma MYB1与海枣(XP_008808341.1)和油棕(XP_010914123.1)的亲缘关系较近。组织特异性研究表明该基因在叶、根和花中的表达量较高。实时荧光定量PCR分析表明Ma MYB1响应干旱、低温、盐和枯萎病菌的侵染等胁迫处理。【结论】Ma MYB1基因在香蕉响应逆境中扮演重要的调控角色。
