桃果肉颜色性状的SSR标记及其在品种中的验证Study on molecular marker linked to flesh color characteristics using SSR in peach and its confirmation in different varieties

摘要(Abstract):
桃果肉颜色是与果品加工密切相关的一种重要经济性状。以桃栽培种瑞光19(白肉)×Summergrand(黄肉)杂交的104株F1后代分离群体为试材,对桃白肉/黄肉的SSR分子标记进行研究,以期为该性状的分子标记辅助育种奠定理论基础。结果表明,筛选的135对引物中有4对引物,CPDCT024、pchgms3、CPPCT026和BPPCT011,在白肉和黄肉单株以及混合基因池间扩增出差异。分析性状与标记的连锁情况表明,CPDCT024与白/黄肉性状遗传距离最近,为7.8cM。试验构建了桃白/黄肉性状的连锁群,根据4个SSR标记与参考图谱T×E和P2175×GN比对,将控制白/黄肉性状的基因定位在参考图谱第1连锁群,对应于桃的第1条染色体。以CPDCT024和参考图谱上另一个与白/黄肉性状连锁距离较近的标记CPPCT026在20个白肉和黄肉品种上验证,CPDCT024检测的准确率为70%,优于CPPCT026。
