桃花芽休眠解除相关上调表达基因的cDNA-AFLP分析Identification of differentially expressed up-regulated genes by cDNA-AFLP approach during dormancy release in peach flower buds

摘要(Abstract):
【目的】桃在冬天为了避免低温伤害而停止生长的发育过程称之为自然休眠。为研究桃花芽休眠解除的分子遗传机制,筛选休眠相关基因。【方法】以中需冷量桃品种‘满天红’为试材,采用cDNA-AFLP技术鉴定花芽内差异表达的基因,并对上调基因进行功能预测。【结果】12对引物共扩增出525条差异TDFs(transcripts-derived fragment),其中与休眠解除相关的上调TDFs 14条,下调的TDFs 18条。对休眠解除后上调的14个差异条带进行克隆测序,经Blast显示其中10个基因分别与泛素连接酶E3、植物组蛋白H3、核糖体蛋白L34、Prohibitin-2蛋白以及半乳糖基转移酶等基因同源性较高。【结论】筛选到的休眠相关基因功能涉及代谢、逆境响应、细胞分化及信号转导,这些基因有助于我们更好的解析桃花芽休眠的机制。
