RFLP在柑桔遗传多样性研究上的应用Study on the Application of RFLP Markers in Assessing Citrus Genetic Diversity

摘要(Abstract):
通过研究摸索出适宜提取柑桔叶片总DNA的方法.利用异源DNA探针——小麦rDNA克隆pTA71,与用四种核酸内切酶EcoR 1、Pst 1、BamH 1及Bgl Ⅱ酶切消化的几种柑桔叶片总DNA杂交,结果表明,pTA71—EcoR 1、pTA71—Pst 1、pTA71—BamH 1、pTAT1—Bgl Ⅱ四种探针一酶组合均可在本地早、宜昌橙、枸橼和金柑等柑桔类型中检测到RFLP差异。利用rDNA的RFLP变异可以从分子水平上研究柑桔遗传多样性,探讨柑桔及其近缘属植物的起源、进化。
