影响苹果赤霉素合成的关键酶基因在新梢和幼嫩种子上的表达分析Expression analysis of genes encoding the key enzymes related to gibberellin biosynthesis in young apple shoots and immature seeds

摘要(Abstract):
【目的】柯巴焦磷酸合成酶(CPS)、贝壳杉烯合成酶(KS)、贝壳杉烯氧化酶(KO)和贝壳杉烯酸氧化酶(KAO)是赤霉素生物合成途径上游的4种关键酶,探讨苹果新梢和幼嫩的种子中编码这4种酶的基因(MdCPS、MdKS、MdKO和MdKAO)在转录水平的表达规律。【方法】以苹果品种‘富士’的新梢和幼嫩的种子为试材,利用实时荧光定量PCR技术,对这4个基因在发育过程中不同时间的转录水平变化进行分析。【结果】这些基因在新梢和幼嫩的种子中均有表达。在新梢中,MdCPS和MdKS、MdKO和MdKAO分别有相似的表达趋势,且在快速生长阶段,MdKO和MdKAO的表达高峰滞后于MdCPS和MdKS。伴随着幼果的迅速膨大,这几个关键酶基因的表达量在幼嫩的种子中急剧增长,且远高于同期的新梢。【结论】这4种关键酶基因的转录水平均与器官的生长速度有关。在生长旺盛的时期,转录水平相对较高。
