龙眼果实ACS和ACO基因克隆及其表达特性Isolation and expression pattern of ACS and ACO genes in longan fruit

摘要(Abstract):
采用RT-PCR和RACE扩增技术相结合的方法从龙眼果实中分离得到ACS和ACO基因,并分别命名为DlACS1和DlACO1。DlACS1蛋白一共编码486个氨基酸,含有7个保守区域和11个不变的氨基残基;DlACO1蛋白一共编码315个氨基酸,含有7个保守区域和9个不变的氨基残基。在龙眼果实发育过程中,果实乙烯释放量在幼果期出现一个小峰,随后逐渐降低,至发育后期维持在较低水平。Northern分析表明,果皮中DlACS1和DlACO1的表达均在果实快速生长期达到最高;在果肉中,DlACS1只在果肉生长初期表达,DlACO1表达随着果肉的生长缓慢降低,在果实成熟期略有升高。此外,DlACS1和DlACO1表达不具有组织特异性,但在各组织中的表达有明显差异。上述结果为研究非跃变型果实的成熟与乙烯之间的关系奠定了基础。
