测定葡萄与葡萄酒总酚的二次回归正交旋转优化设计Optimizing a simple determination method for total phenolics in grape and wine through quadratic regression rotational combination design

摘要(Abstract):
基于不同质量浓度Na2CO3溶液、反应温度和时间对反应体系吸光度影响单因素试验基础上,运用二次回归正交旋转组合设计方法优化反应条件;以微量移液器为工具,建立一种简化、微量测定葡萄与葡萄酒总酚含量的方法。结果表明,建立的回归模型达显著水平,利用优化函数途径进行模拟选优得到最佳反应条件为:0.5 mL样品,0.5mL 1 mol·L-1 Folin-Ciocalteu试剂(F-C试剂)和4.0 mL 52 g·L-1 Na2CO3溶液,在25℃下置102 min后,测765 nm吸光度。相对没食子酸质量浓度在0~0.050 g·L-1内,吸光度和没食子酸浓度有良好的线性关系(R2=0.999 4)。对酿酒葡萄赤霞珠与王朝干红葡萄酒样的加样回收率分别是98.4%和97.8%(n=3),相对标准差(RSD)分别为1.32%和2.01%。该法是一种简化、微量经济测定葡萄与葡萄酒总酚含量的方法,且具有较高的稳定性和准确度。
