利用DGE对黄肉猕猴桃果实着色过程中相关差异基因表达的解析Analysis of differential gene expression during color -turning stages in yellow flesh kiwifruit (Actinidia chinensis) using DGE

摘要(Abstract):
【目的】构建数字基因表达谱(DGE),寻找调控黄肉猕猴桃果实呈色的潜在关键基因。【方法】以中华猕猴桃(Actinidia chinensis Planch)黄肉品种‘金丰’为试材,利用Illimina/Hisseq测序技术构建其果实发育中3个不同转色期DGE文库,以转录组数据为参考数据库,进行GO功能富集以及Pathway功能富集分析。【结果】结果表明,DGE文库构建转色前期、转色期和成熟期分别获得4 724 009,4 565 159和4 626 730个标签。转色期与转色前期之间差异表达基因的个数为2 641个,成熟期与转色前期有差异表达的基因个数是2 671,成熟期与转色期之间有1457个基因有表达差异。通过GO功能富集及Pathway功能富集分析,分别获得了102条叶绿素Unigenes与111条类胡萝卜素Unigenes,其中12条叶绿素Unigenes及14条类胡萝卜素Unigenes在果实的3个不同转色期有显著差异表达(P<0.05)。【结论】黄肉猕猴桃呈色的关键基因可能存在于具有显著差异表达的12条调控叶绿素合成以及14条调控类胡萝卜素合成的Unigenes之中。
