6-BA对抗寒无核葡萄胚挽救的影响Influence of 6-BA on embryo rescue of seedless grape with cold resistance

摘要(Abstract):
【目的】探讨提高抗寒无核葡萄胚挽救育种效率的方法。【方法】以4个种子败育型无核葡萄品种‘火焰无核’‘海王星’‘木星’和‘红宝石无核’为母本,欧山杂种(品种)‘北醇’为父本进行杂交,将杂交胚珠接种于添加不同质量浓度6-BA的MM3+500 mg·L-1水解酪蛋白(CH)+1 mmol·L-1丝氨酸(Ser)培养基上进行培养。【结果】除‘海王星’ב北醇’外,供试的其他杂交组合均在添加6-BA质量浓度为0.5 mg·L-1时,胚珠发育率、萌发率和成苗率最高,差异达到显著水平;当6-BA的质量浓度为1.0 mg·L-1时,对胚挽救效率具一定程度的促进作用或作用不显著;当6-BA的质量浓度为1.5 mg·L-1时,对‘火焰无核’ב北醇’‘木星’ב北醇’‘海王星’ב北醇’杂交后代胚的成苗起到一定程度的抑制作用。【结论】不同的杂交组合对不同质量浓度6-BA的敏感程度不同,0.5 mg·L-16-BA能够提高无核葡萄胚挽救效率;首次选用的抗寒无核品种‘木星’和‘海王星’均可作为无核葡萄胚挽救育种的母本。
