基于genic-SSR标记的MCID法鉴定浙江白沙枇杷地方种质资源Identification of white flesh loquat germplasms of Zhejiang province with MCID strategy using genic-SSR markers

摘要(Abstract):
【目的】建立基于genic-SSR分子标记的鉴定白肉枇杷种质资源的方法,为白肉枇杷资源的品种审定与产业化利用提供依据。【方法】利用10对多态性枇杷genic-SSR引物对新收集的14份浙江地方白沙枇杷种质进行分子鉴定,并应用人工绘制品种鉴别示意图的方法(MCID法)与12个现有白沙枇杷品种建立品种鉴定图(CID)。【结果】仅用5对SSR引物就可将新收集的白沙枇杷新种质与现有品种区分。应用MCID法,依据扩增的多态性核酸片段与对应的引物构建了26份白沙枇杷资源的CID图。从CID图上可找出区分任意2个品种的引物。随机选择各亚组内、亚组间或小组内、小组间的6份种质进行验证,结果与得到的CID图谱一致。聚类结果表明,26个白沙枇杷种质与品种的聚类模式基本符合这些种质的地理起源与遗传背景信息。【结论】基于genic-SSR的MCID法是一种能有效鉴定白沙枇杷种质资源的方法,可应用于枇杷品种鉴定与品种指纹图谱构建。
