桃果实类胡萝卜素测定方法的研究Study the measurement of carotenoids from the peach fruit

摘要(Abstract):
【目的】为了探讨桃果实中类胡萝卜素含量的测定方法,分析桃果实中类胡萝卜素的主要成分及其含量,【方法】以桃品种‘独立’成熟果实为试验材料,采用紫外分光光度计法和高效液相色谱法对不同提取剂、提取方法、色谱柱进行分析比较,筛选出适宜进行桃果实类胡萝卜素定性定量分析的方法。【结果】结果表明,桃果实类胡萝卜素的最佳测定方法是以丙酮为有机提取剂,不经过皂化直接提取,经过0.22 mm有机滤头过滤,采用YMC-C30色谱柱进行测定,流动相A:甲醇,流动相B:甲基叔丁基醚,流动相C:水;流速:1.0 mL·min-1;UV 450 nm;温度为25℃;时间:100 min;进样:20μL;梯度冲洗:0~90 min,81%A/15%B/4%C~6%A/90%B/4%C;92 min,81%A/15%B/4%C。【结论】提取和高效液相色谱检测方法可对桃果实类胡萝卜素进行定性定量分析。
