甜樱桃B类MADS-box基因的分离与序列分析Isolation and sequence analysis of B-class MADS-box gene in sweet cherry

摘要(Abstract):
为研究MADS-box基因在甜樱桃花发育中的作用,以甜樱桃品种拉宾斯为试材,根据多种植物MADS-box基因保守序列设计引物,采用RT-PCR和RACE方法克隆基因cDNA全长,并用半定量PCR法研究基因在不同组织中的表达。克隆得到1个基因全长962 bp,命名为PaMADS2(GenBank登录号:HQ229606),包含1个633 bp的开放阅读框,编码210个氨基酸。序列比对和进化分析表明,PaMADS2与B类的PI基因家族亲缘关系最近。组织特异性分析表明PaMADS2在雄蕊和花瓣中表达。推断所克隆的PaMADS2基因为MADS-box B类家族成员。
