山东牛心柿群体遗传多样性和亲缘关系的SSR分析Genetic diversity and relationship analysis of oriental persimmon (Diospyros kaki) populations in Shandong by SSR

摘要(Abstract):
【目的】从分子水平了解山东牛心柿群体的遗传结构和亲缘关系。【方法】采用SSR分子标记技术对山东省内的8个牛心柿群体共207个个体进行检测,应用POPGENE 1.31软件进行遗传参数分析,采用AREQUIN 2.0软件对群体间和群体内的遗传变异进行分子变异分析,利用TFPGA软件对Nei’s无偏差预期杂合度和基因分化系数进行了统计计算。【结果】10对SSR引物共检测到128个位点,其中105个位点为多态位点,占82%。Nei’s遗传多样性指数(He)在0.153 8~0.247 6,Shannon信息指数(H0)在0.221 7~0.372 1。群体间的遗传变异占总遗传变异的24.17%(P<0.001),群体间的遗传距离(D<0.231 7)、遗传一致度(I>0.783 2)、基因分化系数(φST=0.222 6)和遗传分化系数(FST=0.175 9)均表明各群体间产生了一定程度的遗传分化。【结论】山东牛心柿群体遗传背景丰富,群体间一定程度的遗传分化可能是人为选择压力所引起。
