西洋梨矮化砧木BA-29的组培快繁技术研究Studies on tissue culture and rapid propagation of dwarf rootstock BA-29 of European pear

摘要(Abstract):
以西洋梨矮化砧木BA-29春季茎段为外植体,筛选适宜的消毒时间和诱导、继代增殖与生根培养基,并对试管苗进行了移栽。结果表明,(1)取春季田间嫩茎段为外植体,用0.1%HgCl2消毒5min,污染率可控制在20%以下,外植体成活率超过70%,效果优于消毒3min、7min和10min;以1/2MS+6-BA1mg·L-1+IBA0.1mg·L-1+GA0.2mg·L-1为诱导培养基,增殖倍数较高;(2)适宜继代增殖培养基为MS+6-BA1mg·L-1+IBA0.4mg·L-1+GA0.05mg·L-1,增殖系数可以达到3.8,且芽生长势强,新芽质量好;(3)适宜生根培养基为1/2MS+NAA0.5mg·L-1+IBA0.5mg·L-1,生根率81%,平均生根7.6条,最多达到10条;已生根试管苗炼苗7d后移栽于灭过菌的基质中,30d后移栽成活率超过80%。
