抗生素对中华猕猴桃外植体再生的影响Effects of antibiotics on explant regeneration of Actinidia chinensis

摘要(Abstract):
【目的】为确定中华猕猴桃遗传转化中适宜的抗生素种类和浓度,【方法】以中华猕猴桃‘伏牛95-2’叶片为外植体,研究了抗生素对外植体再生的影响。【结果】抗生素敏感性测定发现,当卡那霉素质量浓度为20 mg.L-1时,愈伤组织及不定芽分化率均为0,因此,20 mg.L-1卡那霉素为适宜的筛选浓度;头孢霉素和羧苄青霉素对中华猕猴桃‘伏牛95-2’叶片分化均有抑制作用,但头孢霉素对叶片分化的抑制作用比羧苄青霉素小,结合抑菌试验,将浓度400 mg.L-1头孢霉素确定为适宜于中华猕猴桃‘伏牛95-2’叶片遗传转化的抑菌抗生素。【结论】卡那霉素20 mg.L-1和头孢霉素400 mg.L-1为中华猕猴桃‘伏牛95-2’遗传转化适宜的抗生素及质量浓度。
